Points to consider when using viral vectors

  • List the genes/siRNA/shRNA being expressed as well as a brief description of the function (known or putative) of the targeted gene/siRNA/shRNA
  • Is expression transient or stable?
  • What is the mechanism of attenuation of the vector?
  • What is the probability of reversion to a virulent phenotype?
  • What is the potential for integration into the host genome?
  • What is the infection tropism (human, animal, plant, etc.) of both the original source virus and the viral vector?
  • What are the factors which make it more or less pathogenic than the source virus?
